I just went through this. Find the grimrock.exe file (usually here: C:\Program Files (x86)\Steam\steamapps\common\Legend of Grimrock). Right click on the file and go to Properties. Then go to Compatibility. In the Compatablilty tab go to "Change High DPI setting". Then make sure Override high DPI scaling behavior is check and then set the drop down menu to "System (Enhanced). This worked for Window 10 using 4k monitors and 1920x1080 resolution. Game graphic looks good, fullscreen, centered and menu is not tiny.

Thank you! This also works for 1440p resolution. The only additional thing I had to do was to set the resolution to fullscreen instead of borderless, otherwise the scaling was broken.

It can be done with dgvoodoo2, the same way I describe in this guide: https://steamhost.cn/steamcommunity_com/sharedfiles/filedetails/?id=2546342859
Confirmed! Plus it works with Grimrock II as well. Still, it does seem to have some peculiar graphical glitches, such as the non-scaled mouse cursor popping up occasionally within the top-left portion of the screen. Any attempt to come out of the game and then go back to it also forces it into windowed mode, which stops the mouse from working at all until you toggle back to full screen (yeah, a minor inconvenience admittedly).

ETA: Speaking of the sequel, I learned only today that the beta version on Steam actually allows for direct modding of the UI. Gave it a try earlier and it is indeed possible to run the game at 4K with the interface scaled at x2. Will definitely be useful for my up and coming recordings of Adrageron's superb Livingrock Adventures.
