No civilian Casulties.
Just something i would like to confirm.

Do i need to kill 150 on single mission or during the game, without hitting civilian and with dual blasters?

No civilian deaths, specifically in the level Longo Two Guns.

Aa so there is a mission called "Longo Two Guns? I though it was a typo.

Yes it's the first mission on Tatooine (Chapter 5).

Does anyone know if an enemy kills a civilian it will ruin the achievement?

What i read, they don't. Only if you kill them. Which can be quite easy since enemies like to hide behind civilians and droids.

In my case, enemies grenades kill many civilians but i still got achievement.

Thank goodness civs killed by enemies don't count. It's almost unavoidable with those grenadiers.

The way those civilians run all over the place, right into your line of fire, drives me mad. You've got an enemy lined up perfectly, you take the shot and suddenly some panicky civilian runs right into it. It's like they want to be killed.

Another question. Do you both achievements in 1 attempt for beating the level?

You mean No civilian Casulties and bounties? You can, but it won't be easy.
So, I know that you need all bounties in 1 playthrough, I'm not talking about that. I'm asking if you need both achievements in 1 playthrough too.

To be specific I barely managed to capture all 15 bounties alive. But I fell short of the kill count I didn't make the 150 but I did manage to have no casualties.

So my question is, if I replay the level for the no casualty achievement, will I have to collect all the bounties again. Or just focus on getting the 150 kills.

Yes, since bounties reset when replaying mission, so you have to collect them again.
Actually, I'm not so sure about that. Doesn't the level keep track of your high score only? What I'm curious about is what makes a high score. Isn't that about capturing bounties? If that's the case, the run when I captured all 15 bounties alive should be the one that kept, I believe. I'm not sure what goes into the score. But if it is indeed about bounties, theoretically I should be able to replay the level, kill 150+ hostile enemies without killing any civilians. In that case the achievement should come out, and as long as I didn't beat my score by collecting any bounties, that should trigger the achievement without overriding my high score. So, those are my thoughts. If not, what exactly goes into "Score" in this game?
