Okay, so I figured it out. Spoilered, though, in case people want to solve the riddle.

It is, indeed, WIlliam, Cyndi's husband; you have to fight him just before he self-destructs, after which he'll become a playable character in Arena Mode.

Ayy! I knew you were going to get it as soon as you made that guess. I really wish I knew how to get the last two. There's probably gonna be some absurdities in finding the other two remaining characters. As for putting it in spoilers, I wouldn't have minded either way personally.

Wait Jilly Lee isn't unlocked in Arena by default when you beat the game?

Does he have a unique moveset? How do you initiate the fight with him?

Just punch him while the cutscene is going on.

It took me maybe two or three tries (because I don't think the flag to unlock him activated, because this game is pretty bugged), but once you defeat him and save, you'll have gotten the character in Arena.

And, uh, he certainly has a unique moveset; The Punch and Kick only go one direction, like pressing punch while facing right makes him kick left, and vice versa.
