My advice is to transcend when you get to a stage where it's becoming too hard to reach the next amount required for another Ancient Soul.
Whilst it can be tempting to keep on struggling upwards (as you've put in a lot of work to get to that point, and you don't want to have to start over again), the best thing to do is to transcend when progress gets too hard, as transcending will make levelling up easier.
As the guide says, "When you transcend for Ancient Souls, you lose any excess hero souls, so I usually transcend just after getting enough for one more Ancient Soul" - i.e. if you're close to getting enough hero souls to get another Ancient Soul, then do it (if not, then go ahead and transcend).
I use pretty much the same principle for ascending and transcending:
When your progress slows down too much, it's time to start over again.
Having autoclickers is great, but doubling your output seems to me to be a priority.
