Intro
We'll be making use of a nifty little tool called "dgVoodoo 2".
Download it here dgVoodoo2[dege.freeweb.hu]
(I would recommend version 2.54, newer ones may have problems like white textures and no save thumbnails).
Unzip the archive and copy "dgVoodooCpl.exe" along with the contents of MS/x86 folder into the Post Mortem main folder.
Tweaking
Now we're ready to tweak some things.
Launch dgVoodooCpl.exe and set the settings like on the images below:

The game will run in 4:3 screen aspect ratio with V-sync on (no tearing), antialiasing and anisotropic filtration. If you want to make the game run in 16:9 but stretched you can change the resolution setting to your preferred resolution.
Done!
All done! Now the game should run on Windows 10 with no problems, just lauch it from Steam as per usual. Note that the same method works for Syberia 1 and 2!

xpollybelle, I fixed this issue by right clicking on the game in the library, go to properties. Under controller I disabled steam input and ran the game successfully.

Sadly, I don't think the game provides any support for controllers. As for the mouse, try to uncheck "capture mouse" option in dgVoodoo. If it doesn't help, try playing aroud with "application controlled fullscreen/windowed state", "disable Alt+Enter to toggle screen state" and try different Output API. I'm sorry that I can't be more specific than this, because for me the mouse works fine and I can't replicate the bug that you're experiencing.

thank you so much, okay it's up and running however my mousepad on my laptop, mouse and controller doesn't work with it, sorry to be a pain! I used to love this game growing up, i'm 29 now and would love to play it again as i have never finished it xxx

xpollybelle, hi! Right click the game in your library and select "Manage" -> "Browse local files".

Mystic Monkey, I wish I could help, but I never had such problems. I'm running it on Windows 10 with the settings from the screenshots and the game works fine. You might try to tweak "Output API" setting and see if it fixes the problem.

Thank you for your help impooving my game, though I have a question. The characters mouth now move in a crazy fashion and I've been noticing bits of white here and there. Is there anyway to fix that?

I also noticed the dgVoodoo v2.72 zip was empty. The other one however seems to work.
