A Complete Tutorial guide to the S4 Vaccine Mod for Project Zomboid. The S4 Vaccine Mod allows you to craft several medical crafting stations and machines used to research and develop Zombie Infection Suppressants, a Cure, as well as other useful items.

The Beginning
This is a tutorial guide for the V4 Vaccine Mod that released on Dec 15th 2024 for B41. I will update this when possible as changes are made. If you are looking for a quick steps process, please refer to the Overview discussion for this mod here. My tutorial is a detailed step by step process that anyone should be able to follow.

This is a first draft, and I may include images later. I have been told that a newer version of this mod is in the works and may hold off on doing that until it is released.

The mod has been updated and this guide is mostly updated to the current version, which I am working on. Though most of the info is currently valid.

To begin, there are a number of first steps one must take. Build the Workbench, and read 3 books. The workbench is located in the craft menu (default button B) in the section called "Vaccine". You'll need carpentry level 2 to build this. The workbench is used to craft the other machines you'll be using later. Crafting the workbench will ask for a number of items and resources. These items will be consumed once the workbench is crafted. So be careful not to lose valuable tools. As for the clothing items listed such as the hard hat and goggles, do not need to be worn. The best way to go about crafting the workbench is to drop all the items on the floor in a single spot, close by to where you want it. Otherwise it'll be too heavy. Stand over the pile of items once you have everything and craft from the menu. The workbench will then be placed in your inventory, and you'll need to "Place" it like placing furniture. It can be moved like regular furniture later

Most of the items required to build the workbench can be found in construction storage containers

At this point, let's go over the 3 books needed. You will need both the following books
  • Basics of Research Equipment
  • Everything About Virology
  • The Basics of Glass Casting
You can find these books in places where you find other skill books. Basically, any bookshelf. Certain places have higher chances of spawning these books. Medical buildings most of all, which you'll most likely be visiting anyway for needed resources

The Basics of Research Equipment book will teach you the recipes for additional work stations made at the workbench. As for the Virology book, it teaches the recipes used at the Chemistry Set station, made at the workbench. The Glass Casting book will teach you glass making recipes to be used at the workbench and the Glass Furnace

Let's go over each equipment, what order they should be built in, and what they do. You will need a minimum of
  • Carpentry 2
  • Metalworking 4
  • Electrical 3
to build everything. You can of course build everything right away if you want to get that out of the way now.

(Note: If you have trouble trying to use the workbench, try to move away from it slightly before right clicking it.)
Gas Furnace
Depending on your situation, the Gas Furnace is the one machine you might not need at all. This machine is used to convert glass into other glass objects you'll be needing, such as test tubes, petri dishes, and syringes. These can be found in places where other medical supplies can be found. Many of these items can be re-used after they have been cleaned with disinfectant. These items are needed to build some of the other machines as well. So if you don't have them, the Gas Furnace should be the first machine you build

The one item this machine does not make, is Pipettes, which you'll be needing later. You can find these in medical locations. These are usually re-usable

(Note: I suspect you might be able to make Pipettes in a future version, as there is currently an unused recipe.)

Once the Gas Furnace is crafted you'll need to "Place" it on a table (or similar) like other appliances (ie a Microwave). To use the Gas Furnace we'll need a couple of items. A Propane Tank, Broken Glass (or glass bottles), a Crucible, Molds, and Tongs. The Crucible, Molds, and Tongs are crafted at the Vaccine Workbench you made earlier. These items are are NOT consumed, so you only need 1 each

With a propane tank in your inventory, right click on the Gas Furnace and "Add Fuel". One unit of propane will add 100 Fuel to the machine. After adding some fuel, right click on it again and Use Gas Furnace. You'll need to add some glass, so click on the "Add Glass" button. This will bring up another UI screen.

Here we can add empty glass bottles or broken glass. Select the little Plus sign "+" or select "All Add Items". Once they have been added, click the "Add Glass" button on this screen, and then close this window.

You should see there is fuel added as well as glass added. Click the "Turn On Gas Furnace" button to start melting the glass down and wait for it to process. This can take some time. The glass should continue to melt down after closing the menu as long as it has fuel. However it will not turn it self off when it is done melting down glass. Don't forget about it and waste fuel

When you are ready to start crafting, select the "View All" button below, or you can select one of the sorted tabs. Follow the recipe guide for what you need crafted.
Autopsy Table / Morgue Table
The Autopsy Table is your starting point in taking Z blood samples. The rest of the machines serve little use if you have no Z blood to use. This table can also be looted from some medical buildings like other furniture. Not all medical buildings have one though. Several can be found at the Louisville Hospital (not advised). Depending on the size of your play space, you might want more than one table, however 1 is enough

How to Use - Once the table is crafted and placed, it's time to bag some zombies (literally). You'll need Sterilized Bags first. To craft a Sterile Bag, you'll need 4 regular garbage bags, and 8 units of Duct Tape (that's 2 full Duct Tape items). These are re-usable, however you'll need Bottles of Disinfectant to clean them. Craft it in the crafting menu (B Button) under Vaccine or right click the garbage bag in your inventory.

After killing a zombie, right click it's corpse and click on "Store in Sterilized Bag". If the option is RED, the corpse has most likely decayed beyond use, and you'll need a fresh zombie kill. Once stored in the bag, you can then transport it. They shouldn't decay once stored in a bag. With a bagged zombie in your inventory, right click on the Autopsy Table and select "Lay Down Corpse"

Now that the corpse has been placed, it's time to take your first sample. You'll need a syringe, a knife, gloves and a mask. I personally like to use a medical mask and surgical gloves along with a scalpel

(Note: Since 1.0 of this mod, you can now take multiple samples depending on your First Aid skill level. Higher skills means more samples. You'll need a syringe per sample.)

Right click on the table with corpse and select Perform Autopsy. You should now have your very first Syringe with Blood Z in it. There are 3 types of zombie blood
  • Blood Z - Normal
  • Blood Z - Good
  • Blood Z - Special
Depending on the type will increase how much research points you can get from them. More on that later. It's time to move on to the next machine... but wait! There's a gross dead zombie on your table. You'll need to clean it up if you want to get more samples. In order to do this, you'll need a garbage bag, a saw, and some Disinfectant, and some cloth like a Sheet or Bath Towel. Time to get rid of this zombie Dexter style. With a saw and garbage bag in your inventory, right click the table to "Remove Corpse". This will create a bag of Medical Waste on the ground. I advise having a nearby garbage bin to throw these bags into and delete. Right click the table once again once the body is removed to clean with disinfectant and cloth. Your table is now clean and ready to be used again

This mod adds a recipe for Disinfectant in the Crafting Menu (B button) under Vaccine. You'll need to find "Large Ethanol Container" to use this recipe. They can typically be found with other medical supplies. If you have trouble finding enough garbage bags, I suggest using a convert plastic bags to garbage bag mod as a partner mod with this one
Chemistry Set
Now that you have a syringe with blood, you'll need the Chemistry Set made and Placed. This one will require Test Tubes and Petri Dishes to construct. This is made at the workbench. Place the Chemistry Set like previous machine. Right click to use.

From here, things start to get more complicated. We'll be coming back to the Chemistry Set often. First we're going to start into Basic Research and understanding. You will notice several recipes listed here tagged with an "[H]" and tagged with a "[Z]"

[H] = Player Blood
[Z] = Zombie Blood

(Note: These are now separated in their own categories, or you can click the View All button.)

For now, we just want to focus on Zombie blood. Find the recipe "[Z]Put Blood in Petri Dish". You will need the syringe with Z blood and an empty clean petri dish. Click the Produce button. You should now have a "Petri Dish[Blood-Z]" item

(Note: Z Blood tagged with "Normal", "Good", and "Special" will lose this tag at this stage. This includes for later when we place blood in test tubes. Although the tag is lost, it appears it is only hidden. It can be found again at the Microscope. Quality of Z blood does not effect anything other than how much research is gained from them)
Microscope
Now that we have something we can use for research, it's time to build the Microscope. Like before, build at the workbench and place. When ready, right click to use.

In the Microscope screen you will see available petri dishes to examine. The View All button should list everything in your inventory that can be used here. The Blood Test section is for examining player blood, while Sample Research is used for Research.

Click the "Dev" button to open the Development Research screen. Here you should see your player name and research points that you have generated on the left side, and things to research on the right side.

Let's take a closer look at these. At the top the first one listed is "Research ZG-D1 Infection Suppressant". To research this you will see in red, Points: 50

Below this is "ZG-D2 Infection Suppressant" which requires Points 150 and SP (Special Points) 10. You'll need the previous one before you can research this one. Next up is "ZG-D3" and finally "ZG-C1". You can probably figure out that the "ZG-C1" is the Cure

Below these are Plasma based research, beginning with "Morphine" In order to research this one, we need [Plasma] Research points. Take note the name difference. The ZG ones need regular research points, and the ones below that, need [Plasma] Research. The other big difference is, the ones that need [Plasma] Research, do not need to be researched in order

We'll come back to the things listed further down the list later. Details on them can be found further down in this guide. For now, we want to focus on the cure and the path to reach it. You can keep the Research window open to see the points change.

Let's examine the blood Z in the Microsoft. Select Sample Research or View All, then select your sample. You will see a display of the blood. Doesn't look too healthy, does it? Click the Research Sample button. You should now have your first research points.

This amount of research however seems small. So let's improve that. Now that you have the basic fundamentals, it's time to step up our research for faster gains. We're going to need extra resources though, but before we move on... what exactly is "ZG-D1"? The ZG-D1 (and the ones after it) are Suppressants. These are used to suppress the zombie infection that a player might be suffering from, if they have been bitten. Basically, they buy you more time before you turn into a zombie
  • ZG-D1 = 24 hour
  • ZG-D2 = 48 hour
  • ZG-D3 = 72 hour
So, how do you check your own blood? With a clean syringe in your inventory, right click on it and select "Blood Collection". After this, go back to the Chemistry Set. This time we want to select "[H]Put Player Blood in Petri Dish". This will give you a "Petri Dish[Blood-H]". Like with the Z blood we just examined, take this to the Microscope. Find it in View All or Blood Test and select it. Hopefully it'll look better than that zombie blood you looked at earlier. Click Perform Blood Test. If all is good, it should say "Not Infected". If you ARE infected, it'll say you are, and what your current infection rate is
Better Research
To get more research, we're going to need a few things
  • Density Gradient Reagent [Histopaque 1077]
  • Density Gradient Reagent [Histopaque 1119]
  • Proteinase K
  • Bottles of Disinfectant
You will also need the following at some point, so keep an eye out for
  • NTP Mix [Nucleotide Triphosphate Mix]
  • Cas9 Enzyme
These materials are harder to come by (other than the disinfectant) and are only typically found where other medical supplies can be found. Although the NTP Mix and Cas9 Enzyme are not required for research, they are required to craft the suppressants and cure. So you'll need them eventually. Depending on your world loot settings, finding these items might be more difficult (or easier). If you're desperate, the Louisville Hospital will most likely have them, even on lower loot settings (I haven't tested that on low loot settings)

Once you have what is listed, we can begin. The starting steps are the same as before. Place a zombie on the table, extract Z blood. Use Chemistry Set. This time we want to select "[Z]Transfer Blood to Test Tube". Next we want to select "[Z]Mix Histopaque 1077/1119 Reagent with Blood"
Centrifuge
If you haven't built the Centrifuge by now, now is the time. Create this at the workbench and then place it. Right click to use it. The Centrifuge can house 6 Test Tubes. This can be useful if you wish to do these steps in bulk

Drag and drop the "Test Tube[Blood=Z + Density Gradient Reagent]" into one of the squares, then turn the machine on. You will now have a "Test Tube[Blood-Z + Density Gradient Reagent, Centrifuged]". Take this to the chemistry station. Select "[Z]Separate Plasma PBMC with Pipette". You will now have 2 items. One "Test Tube [Plasma-Z]" and a "Test Tube [PBMC-Z]"

The PBMC one will be used for Suppression and Cure research, while the Plasma one can be used for the others, such as Morphine. Select the "[Z] Mix Proteinase K Reagent" for both PBMC and Plasma. Go back to the Centrifuge and place both Test Tubes Plasma Z and PBMC Z into it, and turn it on. You should now have Test Tube Plasma Z and PBMC + Proteinase K, Centrifuged. You will need these items again in the future, but for now, we want to get the research from them

To do that, take them back to the Chemistry Set once again. This time we will select "[Z] Wash Plasma with Ethanol to Separate DNA/RNA" and do this as well for the PBMC "[Z]Wash PBMC with Ethanol to Separate DNA/RNA"

We now have Petri Dish with PBMC-Z and Plasma-Z. Take these to the Microscope. Take note how much Research is needed for the Suppressants. Place the PBMC-Z Petri Dish in and click Research Sample. You should see a much larger amount of research. Do this for the Plasma-Z Petri dish as well to start working on the lower row items. More than the simple Z Blood test we did at the start. You should also notice you have gained Special Research as well. But as you can see, you'll need to repeat this a few times to fully unlock research. Once you meet the research requirements, the "Dev" button will become available under the item. Press that to unlock the item.
ZG-D1 Infection Suppressant and Beyond
Once you start unlocking Suppressants, or the Cure, it's time to move on. It's about this time you should have the PCR and the Chromatograph machines made from the workbench, and placed. You'll need them for what's next

Let's make a ZG-D1. For this, we're going to need 2 Test Tubes of PBMC-H. Take note of the "H". That's right, we need YOUR blood now. You should already be familiar with the process. We'll need 2 of these for the suppressant/cure. Take 2 samples of your blood. Place them in a test tube. Mix with 1077/1119 Reagents. We're also going to need 1 PBMC-Z. Take the same steps (test tube & Mix). Once you have your 3 mixed test tubes, take these to the centrifuge. When done, take these back to the Chemistry Set and "Separate Plasma PBMC with Pipette" Do this for both the H and Z versions. You should now have 2 PBMC-H, 2 Plasma-H, and 1 PBMC-Z and Plasma-Z. We wont be needing the Plasma right now, so you can place them in storage. We just want the PBMC

The PBMC-Z needs 1 extra step. Back in the Chemistry Set, we need to select "[Z]Mix Proteinase K Reagent with PBMC" Take this one back to the Centrifuge. This will give you a [PBMC-Z + Proteinase K, Centrifuge]. Time to use the Chromatograph. Drag and drop [PBMC-Z + Proteinase K, Centrifuge] into it and wait for it to process. You should now have a [PBMC-Z Purified for DNA/RNA]

Now it's time to use the PCR machine. You'll need an extra petri dish as well as NTP Mix. We're going to make "ZG-D1 Antibody". When you have all the items ready, click "Create ZG-D1 Antibody" and then Produce.

(Note: I've seen this throw an error, it should still be fine. Do not worry)

You should now have a "Infection Suppressant(ZG-D1) gRNA". Head back to the Chemistry Set and find "Create ZG-D1 Cas9-gRNA Complex". You will need "Cas9 Enzyme" for this in your inventory. When ready, click Produce. Once finished, find "Create ZG-D1 Antibody" and Produce. This recipe will use one of your PBMC-H test tubes.

You now have everything to finally create the ZG-D1 Syringe. When ready select "Create ZG-D1 Syringe". Congrats, you finally have the finished product "Infection Suppressant(ZG-D1)". Keep in mind, this is only a suppressant. If you want the actual Cure, you'll need to continue research and unlock the other Suppressants and finally the Cure. The steps to create the other Suppressant and the Cure are very much the same. Just pay attention to the recipe names. The cure products are labled "ZG-C1"
Plasma Research
So what about that extra unused Plasma? Well, you can turn those into Plasma research. They are also needed to make items from the Plasma Research. Morphine for example, the easiest to unlock, requires Plasma-H. The creation process for Morphine is slightly different, but by now you should have a good idea on how things are made. If you can't find where a certain item is made, make sure to check the PCR. It's easy to forget that machine

But what exactly do all the Plasma Research items do? I will list them and their description. I however have not personally used them yet and can not speak to their validity

Morphine - Lasts 60 minutes. Provides powerful pain relief. Removes pain, fear, stress, and tension during the duration.

Propital - Lasts 60 minutes. Provides powerful pain relief and slight healing effects. Removes pain, fear, stress, and tension while restoring some health during the duration.

eTG-C - eTG-C Regeneration Stimulant. Lasts 20 minutes. Provides powerful healing and slight endurance recovery. Restores a significant amount of health and endurance during the duration.

SJ6 - SJ6 Combat Stimulant. Lasts 120 minutes. Provides powerful endurance recovery. Restores endurance during the duration.

Zargustin - Zargustin Hemostatic. Lasts 30 minutes. Provides powerful hemostatic effects. Stops all bleeding and prevents further bleeding during the duration.

SJ12 - SJ12 Combat Stimulant. Lasts 120 minutes. Removes hunger and thirst during the duration while restoring some calories.
Ending Thoughts
And that's it. Hopefully this has helped someone in some way. If you have any questions or are confused with a step, feel free to ask. If you find a bug, please post that in the bug report on the mod page. I am not the mod author. I will however help solve issues if I can over there.

@Nimbus Unfortunately I haven't been able to test the mod in a multiplayer setting. Do you mean the item it self is not created? I believe originally the mod was coded to require character data to make vaccines. Meaning you needed the human blood sample from the person that was going to be using the end product. I think that part of the mod was put on hold or scrapped, but maybe some left over code is causing an error?

I'm not the creator of the mod, I'd suggest reporting the bug, but it looks like you've already done that. Hopefully he can get back to you on it

Thank you so much for the detailed guide and I'm now at the stage of creating ZG-C1 Treatment gRNA in PCR but I'm having an error and cannot produce anything even ZG-D1 antidote. What does this error means?

ERROR: Multiplayer , 1739783138564> 60,510,810> NetworkZombieMind.set> NetworkZombieMind: goal character is not set.

@BlackThorne Sorry for the delay in response. ZG-D2 should only need 10 SP points, and 150 regular points, by default. PBMC-Z DNA/RNA Petri Dish should reward both points. I recently tested this to make sure there wasn't any bugs, and that all seems to be working as intended

If you are spawning these items into the game through a cheat menu or debug, they will NOT work. Spawned in items have no data points tied to them, which you only get from actually making them through the medical machines. These data points are required to function properly

Items that spawn in the world that are part of a loot table are the only ones that are ok to spawn in through a cheat/debug menu

Also, there was a bug with a previous version of the mod with the ZG research. Make sure your mod is fully up to date

Thanks for this guide, it helped a lot! i got a question, any ideas on why i not getting Special Points when research the PBMC-Z Petri Dish? It just gave me regular points, and for unlock the ZG-D2, it needs 50 Special Points. (sorry for bad english)

@dede loves u You need to mix the blood using the centrifuge with the steps listed. If you're putting the blood just in the petri dish, you get like 5 research out of it. It shouldn't be "zero" though, in which case, that sounds like a bug. Blood right in the petri dish gives minimal research

@Lorien You use it when you get infected. It doesn't give you immunity. I know it's called "Vaccine" but it's more of a cure. You'll need one each time you get infected, so usually good to have a couple on hand for 'just in case'
