(?) Another idea with Misión could be +1 food, +2 faith, +Bonus production to districts and make it give the near districts an adjacency bonus. It would make it a "reverse nubian pyramid" and would make Spain's city planning easier since it will found many citys on coastal tiles, away from rivers or mountains.
TL:DR: Loved the Cultural push to Spain. Makes a lot of sense to switch their Science bias towards it. I would rework "Reducción" 100%. [Encomienda] isn't bad, just hard and situational. [Conquistador] is perfect.

Another fun idea can come with Shipwrecks: many spanish ships take their treasures with them to the depths of the ocean, and it brings an opportunity to push the Cultural side of the nation and make worth of the Harbor great work/relic slot in more modern stages of the game.
[Reducción]: I don't know what a "Reducción" is o.O. The part of bonus production if next to City Center is amazing, but the bonus it provides... feels meh to me.

As an spaniard myself, I think our country was a Culture powerhouse the days of the Colonization... although it was using it as a coercive and violent way of indoctrination. Spain's focus should be there, on Religion - Culture - Domination. Not so much in Science (wtf).
Thats why I love your version of [Phillip II: El Escorial] , although I would replace the 40% bonus to GPP to Culture bonus for Holy Sites equal to its faith and / or a bonus to tourism for great works and relics.
[Conquistador]: is perfect as it is.
[Encomienda]: although the idea isn't bad, maxing this bonus can be a pain to deal with (map can be tricky and spending gov titles to max out trade routes will lead to no-specializated governors until very late in the game.

I tried it out in an immortal difficulty game and found it really fun: The additional yields from trade routes were massive! 14 prod and 28 gold is really just crazy, especially when getting your 7 governors with the help of the natural wonder titles is so easy.
I didn't really make much use of the conquistador UU, because by that point in the game I had pretty much already won - The yields you get from trade routes mean that by the time you have your eighth city (1 capital on continent A and 7 others on continent B), you're pretty much set for the whole game.
I really enjoyed the Reduccion UI, without a doubt the strongest UI in the game. Honestly, just the religion spread along with 1 faith would be good by itself, but the fact that it gives you an extra 3 prod, 3 food and prod to districts is just insane.
Overall I give this a big thumbs up, super fun even if a little OP ;)
