The answer is no, you don't need any DLC installed or enabled.
As soon as I enabled the mod in my 50 week save the boss battle was available and I didn't have any DLC, just vanilla with mods.
I'm on 200 weeks in a play through but not a single Vvlf

This boss is really neat! It sometimes feels like it needs to try a little too hard to force the gimmick but it's otherwise very flavourful and fun to play. I feel like making them all completely immune to movement softlocks too many party comps, however.

It stands out and looks inconsistent due to the absence of the border and the fact that it's larger than other quest icons (at least on the Compact Quest Screen mod).
My ONLY complaint as it stands is how Leon works. I lost one of my lvl 6 Hellions in a retreat, because I didn't have a skill equipped that could target rank 4, and I had already killed Barry so Leon wasn't guarding him. I sustained maybe 15 rounds before just giving up and taking a loss, because I couldn't hit him, which isn't a great feeling. Either he needs to be able to guard Barry's corpse, or Split Glitter needs to deal damage to himself, to help prevent semi-softlocks like that.
Other than that one instance though, every other element of the bossfight works perfectly. Excellent bossfight, 9/10 would die again.
No issues with the Ringmaster class mod (w/ BC trinkets) either.
