I can see sprites being the major sticking point of getting this mod up and running. Good quality sprite work is a difficult skill.
I'm all in for modding and I have quite some experience, only I just started with this game so it will take a while before I start working on mods.
I recommend anyone interested in this concept to have a look at Hero Academy. It's a turn-based strategy game that also contains TF2 classes, it looks really cool. And while it's far from DefQuest's style in terms of gameplay and graphics, it could certainly help with spawning some creative ideas :-)
