Here is my first question, are they both xbox controllers? Because if they're not some games have a hard time difrentiating between different controller inputs.

Second question for if they are both xbox controllers, Did you make sure that: A. the lights on the controllers aren't all messed up, B. if they're wireless plug them in manually because it never works out any other way. The reason I ask part A is because sometimes, (especially with macs but I don't know why you'd have one) the lights on the controller can get pretty wonky as far as recognizing which controller is which. Sometimes if you plug and unplug a controller really fast you can glitch the computer into a state of having all controller slots filled and so if you plug it in again then, yes it might work, but most times you'll result in controlling all of them.

Something else is you might want to check the controls of your game just to see if both player one and 2 are looking at your first controllers input. It can happen sometimes but not often, because the game is supposed to recognize the difference.
