With the Big Shot, a normal headshot
50 - ( 50 x ( 75% x 0.25 = 19% ) )
50 - ( 50 x 0.19)
= 50 - 9
= 41 damage
With the Big Shot, a fully charged headshot
150 - ( 150 x (75% x 0.25 = 19% ) )
150 - ( 150 x 0.19)
= 150 - 28
= 122 damage
