For example Regeneration for Nurgle (Plague of the Crow) is the signature skill and unlock 3rd in the chain. Each of the first 2 skills only allow/take 2 points each, unlike in your pictures where they allow/take 3 each. This means since the signature skill unlocks after 5 points are spent in the chain, and since a maximum of 4 points can be spent on the chain, it can never unlock and can never be chosen.

Slaughterpriests are the demagogues of Khorne's
wrathful will, able to control the flows of blood
coursing through the veins of warriors. They are dark
prophets who receive gory visions of violence and
death from their god. Through dread invocations a
Slaughterpriest can also boil the blood surging inside
of their enemies, causing cries of agony as eyes burst in
fountains of red fluid, or whip their allies into a savage
battle-frenzy.
