What mods seem the most "vanilla" to you?
Just wondering. I'm guessing most QoL mods and maybe some utility mods. But I would love to hear opinions on this.

I only dabble with characters, so my only answer is the templar. If you swapped him with defect, it'd be hard to tell which concept came first.

Most like the original game? Probably the Firefly. It's a very tightly designed character.

Most in the spirit of the original game? Replay the Spire

From what I gathered, it is quite evident which mods are 'vanilla' since there a basically these categories:

Additional Features and Functionalities which one can pick and choose. <- Obviously as vanilla as they seem to be to one. There are plenty I could recommend, whether it is a corruption mechanic (Corrupt the Spire), questing to climb infinitely (Infinite Spire), Wandering Minibosses or Battle Towers.

Original Content Additions (Enemies, Relics and Cards): I found most of them quite vanilla, however two, after all this time, have glaring 'beta'-art with either a single symbols (Replay the Spire) or 'Beta' written in MS Paint (Hubris). As much as I like the gameplay of these two...

Original Characters: Popular+Updated are usually the best, look at the comments to see if they still work well.

Cross-Over (non original): Can be as seamless but can be quite glaring

Chinese: After more or less (less) extensive testing, I found that the big mods sure put in a lot of everything but I found their everything gameplay related 'over the top'. I'm not really willing to test further than what I did (or can, the first elites do 40+ damage on turn 3) but I suppose that they are standalone. One basically has to use the added characters which acquire primarily the added relics (so no relic pool bloat) to last through the events and encounters.
